1. HF neutralization test

GluCA™ Gel 2.5% (pH 5.71)
GluCA™ Gel 2.5% 10g + 10% HF 1g (pH 3.57)
  • ① pH Change: When 1 gram of 10% HF (pH < 1, Strong Acid) added into 10 gram of GluCA™ Gel 2.5%, pH of 10% HF increase from
    < 1 to 3.57.
  • ② Precipitation: Originally, the outlook of GluCA™ Gel 2.5% is transparent gel. After 1 gram of 10% HF added into 10 gram of GluCA™
    Gel 2.5%, however, the outlook changed to white opaque gel right away due to the precipitation of CaF2 complex (Ca2+ + F- → CaF2).

3. Artificial 3D human skin tissue model test

1) EpiDerm™ tissue model (MatTek Co., MA, USA)

  • ① Normal human 3D (NHu-3D) model of epidermal tissue
  • ② Produced from normal epithelial keratinocytes
  • ③ In Vivo-like lipid profile
  • ④ transdermal drug delivery/percutaneous absorption

2) Skin irritation & corrosion test

  • EpiDerm™ system consists of normal, human-derived epidermal keratinocytes (NHEK) which have been cultured to form a multilayered, highly differentiated model of the human epidermis. These "ready-to-use" tissues, also known generically as Normal Human-3D (NHu-3D), are cultured on specially prepared cell culture inserts using serum free medium, attain levels of differentiation on the cutting edge of in vitro skin technology. Ultrastructurally, the EpiDerm Skin Model closely parallels human skin, thus providing a useful in vitro means to assess dermal irritancy and toxicology.

4. Mouse model test

1) Measurement of calcium ion concentration within mouse blood plasma: analyzed and compared calcium ion concentration of mouse
     blood plasma between HF-treated mouses with and without(comparison group) GluCA™ Gel 2.5% application.
2) Clinical severity score test: After GluCA™ Gel 2.5% applied on the surface of HF-treated mouse skins, clinical severity scores are checked
     as the base of 0 (none), 1 (mild), 2 (moderate), 3 (severe), and compared to reference image of no-application (comparison group).

3) Immunohistochemical staining test: Using target antigen related to wound healing, wound healing of HF burned skin can be evaluated to
     form immunoglobulin complex, and staining the complex and check the revelation of the index related to wound healing of HF damaged
     skin.
